SS-31 (Elamipretide / MTP-131 / Bendavia) is a synthetic mitochondrial-targeting tetrapeptide developed by the Hazel Szeto laboratory and licensed to Stealth BioTherapeutics. The peptide consists of an aromatic-cationic sequence (D-Arg-2′,6′-dimethylTyr-Lys-Phe-NH₂) designed to selectively concentrate in the inner mitochondrial membrane through electrostatic attraction to the negatively charged headgroup of cardiolipin — a unique mitochondrial phospholipid critical for organization of electron transport chain supercomplexes.
