Sermorelin (also known as GHRH 1-29 or GRF 1-29) is a synthetic 29-amino-acid peptide corresponding to the N-terminal active fragment of human growth hormone–releasing hormone (GHRH). Originally developed by Lance Frohman's group and characterized in the 1980s, sermorelin represents the minimum sequence required for full biological activity at the GHRH receptor — the C-terminal portion of native 44-amino-acid GHRH is dispensable for receptor agonism. Sermorelin functions as a selective GHRH receptor agonist that stimulates pulsatile endogenous growth hormone release from anterior pituitary somatotropes, with downstream hepatic IGF-1 production. The compound was historically FDA-approved as Geref® (Serono) in 1997 for pediatric GH deficiency before being discontinued commercially. Sermorelin has been investigated extensively in preclinical research for GHRH receptor pharmacology, somatotropic axis signaling, and age-related GH decline research models.

Benefits (Research Focus)

GHRH receptor agonism — studied as the prototypical natural GHRH analog for receptor pharmacology research

Pulsatile GH release — investigated for physiologic-pattern growth hormone release from pituitary somatotropes

IGF-1 axis signaling — explored for downstream hepatic IGF-1 production and somatotropic endpoints

Age-related GH decline research — examined in aged rodent models of somatotropic axis function

Combination pharmacology — researched alongside GHS-R agonists (e.g., ipamorelin, MK-677) for paired pathway studies

What Researchers Look At

• Plasma GH and IGF-1 dynamics following sermorelin administration

• GHRH receptor binding affinity and pituitary somatotrope responses

• Comparative pharmacology versus modified GHRH analogs (CJC-1295 No DAC, tesamorelin)

• DPP-IV cleavage kinetics and short half-life pharmacokinetics

• Age-associated changes in pituitary GH responsiveness in research models
