Kisspeptin-10 (also known as KP-10 or Metastin 45-54) is the 10-amino-acid C-terminal active fragment of the kisspeptin family of peptides, encoded by the KISS1 gene and originally characterized as the natural ligand for the GPR54 / Kiss1R receptor. Kisspeptin signaling is now recognized as the master upstream regulator of the hypothalamic-pituitary-gonadal (HPG) axis — kisspeptin neurons in the arcuate nucleus and anteroventral periventricular nucleus directly stimulate hypothalamic GnRH (gonadotropin-releasing hormone) neurons, which in turn control pituitary LH and FSH release. The C-terminal -RFG-NH₂ amidation pattern is essential for biological activity, making KP-10 the minimum active sequence with full agonist potency at Kiss1R. The compound is investigated extensively in preclinical and clinical research for reproductive endocrinology, HPG axis pharmacology, puberty timing, hypothalamic neuroendocrine signaling, and Kiss1R receptor characterization.
• Kiss1R / GPR54 agonism — studied as the natural ligand for the kisspeptin receptor (formerly orphan GPR54)
• HPG axis signaling — investigated for upstream regulation of hypothalamic GnRH neurons
• Gonadotropin release — explored for downstream LH and FSH pituitary response endpoints
• Hypothalamic neuroendocrinology — examined for arcuate and AVPV nucleus kisspeptin neuron pharmacology
• Reproductive research — researched for HPG axis function in preclinical reproductive endocrinology models
• Kiss1R / GPR54 receptor binding affinity and downstream signaling cascades
• Plasma LH and FSH dynamics following Kisspeptin-10 administration
• Hypothalamic GnRH neuron activation patterns and pulse generator function
• Kisspeptin neuron populations in arcuate (Kiss1/NKB/Dyn) and AVPV nuclei
• Comparative pharmacology versus full-length kisspeptin and other KP fragments (KP-13, KP-14, KP-54)
