FOXO4-DRI is a synthetic D-retro-inverso peptide designed to selectively disrupt the FOXO4–p53 protein-protein interaction. The compound was developed by Peter de Keizer and colleagues at Erasmus University Medical Center (Rotterdam) and published in a landmark 2017 Cell paper (Baar et al., Cell 169:132-147) demonstrating selective induction of apoptosis in cellular senescence research models. The "D-retro-inverso" design replaces all L-amino acids with D-amino acids and reverses the sequence — a structural strategy that preserves the side-chain topology of the original FOXO4 binding region while making the peptide resistant to proteolytic degradation, dramatically extending plasma half-life compared to the natural L-form.
